EHMT2-mediated R-loop formation promotes the malignant progression of prostate cancer via activating Aurora B. [PDF]

open access: yesClin Transl Med
Euchromatic histone lysine methyltransferase 2 (EHMT2) is overexpressed in advanced prostate cancer, restraining catastrophic chromosomal instability (CIN) and enhancing cell fitness. EHMT2 functions via the centromeric R‐loop‐driven ATR–CHK1–Aurora B pathway to promote chromosomal stability.

Age‐dependent aneuploidy in mammalian oocytes instigated at the second meiotic division

open access: yesAging Cell, Volume 21, Issue 7, July 2022., 2022
Ageing severely affects the chromosome segregation process in human oocytes resulting in aneuploidy, infertility and developmental disorders. We have studied the second meiotic division in oocytes from aged mice and established that post‐metaphase II processes have critical impact on age‐dependent aneuploidy in mammalian eggs.

Novel cis‐Pt(II) Complexes with Alkylpyrazole Ligands: Synthesis, Characterization, and Unusual Mode of Anticancer Action

open access: yesBioinorganic Chemistry and Applications, Volume 2022, Issue 1, 2022., 2022
One concept of improving anticancer effects of conventional platinum‐based antitumor drugs consists of conjugating these compounds with other biologically (antitumor) active agents, acting by a different mechanism. Here, we present synthesis, physicochemical characterization, biological effects, and mechanisms of action of four new analogs of ...

Age‐dependent integrity of the meiotic spindle assembly checkpoint in females requires Aurora kinase B

open access: yesAging Cell, Volume 20, Issue 11, November 2021., 2021
AURKB in needed to preserve the female reproductive lifespan likely by protecting oocytes from oxidative stress. We show that Aurkb KO oocytes prematurely accumulate ROS during maternal aging. Prophase‐arrested oocytes from older females have reduced expression of SAC signaling components, causing a weakened SAC response during meiosis I.

Interaction of the mitotic inhibitor monastrol with human kinesin Eg5 [PDF]

open access: yes, 2003
The microtubule-dependent kinesin-like protein Eg5 from Homo sapiens is involved in the assembly of the mitotic spindle. It shows a three-domain structure with an N-terminal motor domain, a central coiled coil, and a C-terminal tail domain. In vivo HsEg5

Inhibitor library screening identifies ispinesib as a new potential chemotherapeutic agent for pancreatic cancers

open access: yesCancer Science, Volume 112, Issue 11, Page 4641-4654, November 2021., 2021
Ispinesib mediated anti–proliferative effects and significantly reduced the growth of Eg5‐positive pancreatic cancer in a patient‐derived xenograft mouse model. Moreover, Eg5‐positivity was associated with poorer clinical outcomes in pancreatic cancer patients.
