Ryanodine Receptor 1 Is Dispensable for CD4+ T-Cell Differentiation and Effector Function in Intestinal Inflammation Models. [PDF]

open access: yesEur J Immunol
Ryr1 deletion reduces Ca2+ microdomains and delays global Ca2+ signaling in naïve CD4+ T cells but does not impair their activation, proliferation, or differentiation in vitro. Despite altered Ca2+ signaling, Ryr1 expression in CD4+ T cells was dispensable for their pathogenicity in murine models of intestinal inflammation.

The Na+/K+-ATPase β1 Subunit is a Kidney ADP-Ribosyl Cyclase. [PDF]

open access: yesFASEB J
This study highlights that the β1 subunit of Na, K‐ATPase, known as Atp1b1, functions as an ADP‐ribosyl cyclase in rat kidney (kARC). We found that reducing Atp1b1 expression in mouse mesangial cells suppressed angiotensin II (AngII)–induced synthesis of cyclic ADP‐ribose (cADPR) and the associated sustained Ca2+ signaling.

MASTER-NAADP: a membrane permeable precursor of the Ca2+ mobilizing second messenger NAADP [PDF]

open access: yesNature Communications
Upon stimulation of membrane receptors, nicotinic acid adenine dinucleotide phosphate (NAADP) is formed as second messenger within seconds and evokes Ca2+ signaling in many different cell types.

Nicotinic Acid Adenine Dinucleotide Phosphate (NAADP)-Mediated Calcium Signaling Is Active in Memory CD4+ T Cells [PDF]

open access: yesMolecules
Nicotinic acid adenine dinucleotide phosphate (NAADP), identified as one of the most potent calcium-mobilizing second messengers, has been studied in different eukaryotic cell types, including lymphocytes.

Two-pore channel regulators - Who is in control? [PDF]

open access: yesFrontiers in Physiology
Two-pore channels (TPCs) are adenine nucleotide and phosphoinositide regulated cation channels. NAADP activates and ATP blocks TPCs, while the endolysosomal phosphoinositide PI(3,5)P2 activates TPCs.

Tetrandrine regulates NAADP-mediated calcium signaling through a LIMP-2-dependent and sphingosine-mediated mechanism [PDF]

open access: yesNature Communications
Tetrandrine (Tet) is a potent inhibitor of Ebola virus replication by blocking NAADP-dependent calcium release through endolysosomal two-pore channels (TPCs) and a moderately potent anti-tumor agent.

Ionic Mechanisms of Two-Pore Channel Regulation of Vesicle Trafficking [PDF]

open access: yesCells
The endolysosomal system plays a pivotal role in cellular function. Before reaching lysosomes for degradation, the endocytosed cargoes are sorted at various stages of endosomal trafficking for recycling and/or rerouting.

Metabotropic Glutamate Receptor-Dependent Synaptic Plasticity in Age-Related Neurodegenerative Disorders. [PDF]

open access: yesNeural Plast
Synaptic plasticity is a fundamental property of the nervous system that underpins learning, memory, and adaptive behavior across the lifespan. Disruption of plasticity mechanisms is increasingly recognized as a unifying feature of age‐related neurodegenerative and neuropsychiatric disorders.

Two-pore channel-2 controls calmodulin-dependent STIM1 inactivation [PDF]

open access: yesNature Communications
Lysosomal two-pore channels (TPC) trigger Ca2+ release from the endoplasmic reticulum (ER). The ensuing ER Ca2+ depletion activates STIM1-gated store-operated Ca2+ entry (SOCE) channels that sustain Ca2+ signals regulating fundamental cellular processes.

JPT2 Affects Trophoblast Functions and Macrophage Polarization and Metabolism, and Acts as a Potential Therapeutic Target for Recurrent Spontaneous Abortion. [PDF]

open access: yesAdv Sci (Weinh)
Dysregulated Jupiter microtubule‐associated homolog 2 (JPT2) at the maternal–fetal interface leads to recurrent spontaneous abortion (RSA) development. The present study shows that JPT2 deficiency inhibits trophoblast function. Furthermore, the lack of JPT2 in trophoblasts contributes to M1 macrophage polarization by promoting the accumulation of ...
