P7C3 Compounds as Targeted Mitochondrial Therapeutics for Brain Disorders

open access: yesCNS Neuroscience &Therapeutics, Volume 32, Issue 9, September 2026.
P7C3 activates NAMPT, the rate‐limiting enzyme in the NAD+ salvage pathway, elevating NAD+ levels in neurons. This engages SIRT1‐dependent nuclear signaling and SIRT3‐dependent mitochondrial regulation, enhancing mitochondrial quality control, reducing oxidative stress, and promoting neuronal survival.

Oral Administration of Nicotinamide Mononucleotide Increases Nicotinamide Adenine Dinucleotide Level in an Animal Brain

open access: yes, 2022
As a redox-sensitive coenzyme, nicotinamide adenine dinucleotide (NAD+) plays a central role in cellular energy metabolism and homeostasis. Low NAD+ levels are linked to multiple disease states, including age-related diseases, such as metabolic and ...

Safety and efficacy of long-term nicotinamide mononucleotide supplementation on metabolism, sleep, and nicotinamide adenine dinucleotide biosynthesis in healthy, middle-aged Japanese men

open access: yesEndocrine Journal
Obesity and aging are major risk factors for several life-threatening diseases. Accumulating evidence from both rodents and humans suggests that the levels of nicotinamide adenine dinucleotide (NAD+), a regulator of many biological processes, declines in

A nonrandomized study of single oral supplementation within the daily tolerable upper level of nicotinamide affects blood nicotinamide and NAD+ levels in healthy subjects

open access: yesTranslational Medicine of Aging, 2020
A decrease in nicotinamide adenine dinucleotide (NAD+) levels is a hallmark of aging in multiple organisms, including humans. We report a human clinical trial designed to assess the efficacy of single-dose supplementation of nicotinamide, a form of ...

Exogenous nicotinamide mononucleotide, NAMPT-catalyzed reaction end product, reverts the inhibitory effects of APO866.

open access: yes, 2013
Mouse PEC were incubated in the presence or absence of 200 nM APO866 and 10 mM nicotinamide mononucleotide (NMN). Cells were further stimulated with LPS and intracellular NAD levels and TNFα and IL-6 secretion were determined.

SLC29A1 and SLC29A2 are human nicotinamide cell membrane transporters

open access: yesNature Communications
Nicotinamide (NAM), a main precursor of NAD+, is essential for cellular fuel respiration, energy production, and other cellular processes. Transporters for other precursors of NAD+ such as nicotinic acid and nicotinamide mononucleotide (NMN) have been ...

Role and Potential Mechanisms of Nicotinamide Mononucleotide in Aging

open access: yesAging and disease
Nicotinamide adenine dinucleotide (NAD+) has recently attracted much attention due to its role in aging and lifespan extension. NAD+ directly and indirectly affects many cellular processes, including metabolic pathways, DNA repair, and immune cell activities. These mechanisms are critical for maintaining cellular homeostasis.

The function of nicotinamide phosphoribosyl transferase (NAMPT) and its role in diseases

open access: yesFrontiers in Molecular Biosciences
Nicotinamide phosphoribosyl transferase (NAMPT) is a rate-limiting enzyme in the mammalian nicotinamide adenine dinucleotide (NAD) salvage pathway, and plays a vital role in the regulation of cell metabolic activity, reprogramming, aging and apoptosis ...
