A sulfur-aromatic gate latch is essential for opening of the Orai1 channel pore

open access: yeseLife, 2020
Sulfur-aromatic interactions occur in the majority of protein structures, yet little is known about their functional roles in ion channels. Here, we describe a novel molecular motif, the M101 gate latch, which is essential for gating of human Orai1 ...

Endoplasmic Reticulum Geometry Dictates Neuronal Bursting via Calcium Store Refill Rates and Exposes Selective Neuronal Vulnerability

open access: yesAdvanced Science, Volume 13, Issue 36, 29 June 2026.
The ER's continuous tubular network is maintained by ER‐shaping proteins whose mutation or dysregulation contributes to neurodegenerative diseases. Here, we show that ER morphology sets the speed of Ca2+ store replenishment between firing events. Disrupting ER continuity slows intra‐ER Ca2+ redistribution from extracellular refill (SOCE) sites, driving

Ryanodine Receptor Ca2+ Leak‐Induced Redistribution of Ca2+ in Dystrophic mdx Mouse Muscle

open access: yesActa Physiologica, Volume 242, Issue 6, June 2026.
ABSTRACT Aim The dystrophic mdx mouse is a widely used model of Duchenne muscular dystrophy. Altered Ca2+ handling is a key feature, including increased Ca2+ leak through the ryanodine receptor (RyR1's), the primary Ca2+ release channel in skeletal muscle. Such leak has important downstream consequences for intracellular Ca2+ homeostasis.

Up-regulation of Orai1 in mdx muscle.

open access: yes, 2012
(A) mRNA expression levels of Orai1 and STIM1 in gastrocnemius muscles from wt C57BL/10ScSnJ and dystrophic C57BL/10ScSn-Dmdmdx/J mice were detected by real-time PCR.

Calcium Buffering in Astrocytes and Its Relevance for Experimental Data Interpretation and Computational Modeling

open access: yesJournal of Neurochemistry, Volume 170, Issue 6, June 2026.
Components and properties of Ca2+ buffers in astrocytes, which should be considered for data interpretation and in computational modeling of astrocyte Ca2+ activity. ABSTRACT Astrocytic Ca2+ signaling is essential for maintaining physiological brain function, including the modulation of synaptic transmission, neurovascular coupling, and ion homeostasis.

Expressed protein ligation for a large dimeric protein [PDF]

open access: yes, 2011
Expressed protein ligation (EPL) is a protein engineering tool for post-translational ligation of protein or peptide fragments. This technique allows modification of specific parts of proteins, opening possibilities for incorporating probes for ...

Amplification of CRAC current by STIM1 and CRACM1 (Orai1). [PDF]

open access: yes, 2006
Depletion of intracellular calcium stores activates store-operated calcium entry across the plasma membrane in many cells. STIM1, the putative calcium sensor in the endoplasmic reticulum, and the calcium release-activated calcium (CRAC) modulator CRACM1 (
