Pregnenolone and pregnenolone sulfate, alone and with ethanol, in mice on the plus-maze

Pharmacology Biochemistry and Behavior, 1994
The neurosteroids pregnenolone and pregnenolone sulfate were tested for anxiogenic/anxiolytic effects in mice on the elevated plus-maze. Pregnenolone in a dose of 0.01 micrograms/kg increased motor activity and caused an anxiogenic response, i.e., a decreased number of entries onto the open arms of the plus-maze.

Metabolism of Pregnenolone and Pregnenolone Sulphate in Benign Essential Hypertension

Endocrine Research Communications, 1974
Patients with benign essential hypertension excreted in their urine less of the radioactive label associated with the injected tracer 3H-pregnenolone sulphate than control subjects, but more of the label associated with the 14C-pregnenolone tracer. In both groups, there was no difference in the urinary production rate of pregnenolone.

The quantitative determination of pregnenolone and pregnenolone acetate by infrared spectrophotometry

Journal of the American Pharmaceutical Association (Scientific ed.), 1950
An infrared spectrophotometric method is described for the quantitative determination of pregnenolone and its acetate in pharmaceutical preparations. Optical density measurements are made at several intense absorption peaks, and the concentration is calculated independently from each measurement.

Measurement of pregnenolone in blood

Steroids, 1970
Abstract A radio-ligand assay for the measurement of pregnenolone in 5 ml of plasma has been devised. Pregnenolone is converted to progesterone enzymatically and measured as progesterone. The method has low blank value and high sensitivity (0.3 ng). Plasma concentrations of pregnenolone were (means ± SEM, ng/ml): men, 0.83 ± 0.08; women, follicular ...

Pregnenolone in Rheumatoid Arthritis

New England Journal of Medicine, 1951
PREGNENOLONE (Δ 5-pregnen-3βol-20-one) has recently been recommended as effective in the treatment of rheumatoid arthritis.1 2 3 4 5 Since Cortisone (17 hydroxy-11 dehydrocorticosterone) and ACTH (adrenocorticotrophic hormone) are scarce, expensive and not free from side effects, interest in pregnenolone is growing.

Biosynthesis of Pregnenolone

1971
Publisher Summary Pregnenolone is the C21 steroid closest in structure to cholesterol, which is the most abundant sterol in the Eutheria. C21 steroid is by far the major precursor of all the steroid hormones in the adrenal gland, ovary, and testis.

Pregnenolone production in pregnancy

American Journal of Obstetrics and Gynecology, 1971
Abstract The blood metabolic clearance rate of pregnenolone was 2,950 L. per day at 11 to 12 weeks' gestation, 2,670 ± 389 (S.E.) L. per day between 33 and 36 weeks' gestation, and 4,250 ± 502 (S.E.) L. per day between 37 and 39 weeks' gestation. Thirty-three per cent of circulating pregnenolone was associated with the blood cells. In five subjects in

Radioimmunoassay of Plasma Pregnenolone

The Journal of Clinical Endocrinology & Metabolism, 1973
ABSTRACT A radioimmunoassay for the measurement of plasma unconjugated pregnenolone (5Δ-P) is described. The 3-hemisuccinate of 5Δ-P was coupled to human serum albumin using carbodiimide. One adult ewe was immunized with 2 mg of this conjugate. The antiserum obtained after 7 months of immunization was used in the assay. One to two ml of plasma or serum,

Forgetting in obesity: The pregnenolone link

Cell Metabolism, 2022
Cognitive dysfunction is often diagnosed in people with obesity and associated metabolic disorders. In the latest issue of Cell Metabolism, Ramírez et al. highlight an impaired production of the neurosteroid pregnenolone in the hypothalamus as a mechanism for obesity-induced cognitive impairment in both rodent models and patients with obesity.

PLASMA PREGNENOLONE AND 17-OH-PREGNENOLONE IN HIRSUTE AMENORRHOEIC PATIENTS

Acta Endocrinologica, 1978
ABSTRACT This study was designed (a) to examine the involvement of the Δ5-pregnenes, pregnenolone and 17-OH-pregnenolone in the abnormal steroidogenesis present in hirsute patients; and (b) to examine the possibility that suppressed adrenal 21-hydroxylase activity may frequently underlie the abnormal steroidogenesis in hirsute women.
