EFFECTS OF IL-4 UPON THE ACTIVITY OF STAT6 TRANSCRIPTION FACTOR IN PERIPHERAL BLOOD LYMPHOCYTES IN BRONCHIAL ASTHMA [PDF]
. The aim of the study is to specify the levels of STAT6, and phospho-STAT6 under the influence of IL-4 in patients with bronchial asthma (BA). The samples from ten healthy controls and thirty-three BA patients with allergic and non-allergic clinical ...

Genome-wide Profiling of Interleukin-4 and STAT6 Transcription Factor Regulation of Human Th2 Cell Programming [PDF]
Dissecting the molecular mechanisms by which T helper (Th) cells differentiate to effector Th2 cells is important for understanding the pathogenesis of immune-mediated diseases, such as asthma and allergy. Because the STAT6 transcription factor is an upstream mediator required for interleukin-4 (IL-4)-induced Th2 cell differentiation, its targets ...

Herpesvirus saimiri (HVS), a T-lymphotropic monkey herpesvirus, induces fulminant T-cell lymphoma in non-natural primate hosts. In addition, it can immortalize human T-cellsin vitro. HVS tyrosine kinase-interacting protein (Tip) is an essential viral gene required for T-cell transformation bothin vitroandin vivo.

LPS induces the interaction of a transcription factor, LPS-induced TNF-α factor, and STAT6(B) with effects on multiple cytokines [PDF]
TNF-α is a pivotal cytokine whose overproduction can be lethal. Previously, we identified a transcription factor, LPS-induced TNF-α factor (LITAF), that regulates TNF-α transcription. We now report the discovery and characterization of a regulatory cofactor that we call signal transducer and activator of transcription (STAT) 6(B) because of its ...

GM-CSF Promotes Neutrophil PD-L1 Expression Through the VEGFR-2/STAT6/CSF2 Signaling Axis in Oral Squamous Cell Carcinoma. [PDF]
VEGFR‐2 signaling in OSCC activates Src–STAT6‐dependent CSF2 transcription, driving tumor‐derived GM‐CSF secretion. GM‐CSF programs neutrophils to express PD‐L1 through STAT5–mTOR/S6K signaling, suppressing cytotoxic CD8+ T cells. This pathway reveals a tumor–neutrophil immune checkpoint circuit that limits anti‐PD‐1 responsiveness in OSCC.

IL-6 augments IL-4-induced polarization of primary human macrophages through synergy of STAT3, STAT6 and BATF transcription factors [PDF]
Macrophages in the tumor microenvironment respond to complex cytokine signals. How these responses shape the phenotype of tumor-associated macrophages (TAMs) is incompletely understood. Here we explored how cytokines of the tumor milieu, interleukin (IL)-

4-1BBL Suppresses Anti-Inflammatory Responses by Regulating Metabolic Reprogramming of Macrophages. [PDF]
4‐1BB ligand (4‐1BBL) is known to promote sustained pro‐inflammatory responses in macrophages. This study demonstrates that 4‐1BBL acts as a negative regulator of anti‐inflammatory macrophage responses. Genetic deletion or pharmacological inhibition of 4‐1BBL significantly enhanced the phosphorylation of IL‐4R‐proximal JAK1 and STAT6, elevating the ...

Transcription Factors STAT6 and KLF4 Implement Macrophage Polarization via the Dual Catalytic Powers of MCPIP [PDF]
Abstract Macrophage polarization plays a critical role in tissue homeostasis, disease pathogenesis, and inflammation and its resolution. IL-4–induced macrophage polarization involves induction of STAT6 and Krüppel-like factor 4 (KLF4), which induce each other and promote M2 polarization. However, how these transcription factors implement

T helper type 2 inflammatory disease in the absence of interleukin 4 and transcription factor STAT6 [PDF]
An important signaling pathway for the differentiation of T helper type 2 (TH2) cells from uncommitted CD4 T cell precursors is activation of the STAT6 transcription factor by interleukin 4 (IL-4). The protooncogene BCL-6 is also involved in TH2 differentiation, as BCL-6 −/− mice develop an inflammation of the heart and lungs ...
