Phenome-wide association studies on a quantitative trait: application to TPMT enzyme activity and thiopurine therapy in pharmacogenomics. [PDF]

open access: yesPLoS Computational Biology, 2013
Phenome-Wide Association Studies (PheWAS) investigate whether genetic polymorphisms associated with a phenotype are also associated with other diagnoses.

Thiopurine S‐methyltransferase genetic polymorphism in the Thai population [PDF]

open access: yesBritish Journal of Clinical Pharmacology, 2004
AimsTo determine the incidence of the thiopurine S‐methyltransferase (TPMT) genetic polymorphism in the Thai population.MethodsGenomic DNAs were isolated from peripheral blood leucocytes of 200 healthy Thais. The frequencies of five allelic variants of the TPMT gene, TPMT*2, *3A, *3B, *3C and *6 were determined using allele specific polymerase chain ...

Personalization of thiopurine therapy: Current recommendations and future perspectives

open access: yesActa Pharmaceutica
Despite great therapeutic advances in the field of biologics, small synthetic molecules such as thiopurines, including azathioprine, mercaptopurine, and thioguanine, remain an important therapeutic pillar in the treatment of inflammatory bowel disease ...

Frequency of thiopurine S-methyltransferase gene variations in Turkish children with acute leukemia

open access: yesThe Turkish Journal of Pediatrics, 2018
In this study we aim to determine the genotype distribution and allele frequencies of common TPMT (*2, *3A, *3B and *3C) polymorphisms in Turkish children with acute leukemia.

Role of Oxidative Stress Mediated by Glutathione-S-transferase in Thiopurines' Toxic Effects [PDF]

open access: yes, 2015
Azathioprine (AZA), 6-mercaptopurine (6-MP), and 6-thioguanine (6-TG) are antimetabolite drugs, widely used as immunosuppressants and anticancer agents.

Direct evidence of singlet molecular oxygen [O2 (1Δg)] production from UVA excited 6‐thioguanine

open access: yesPhotochemistry and Photobiology, EarlyView.
6‐Thioguanine (6‐TGua) is incorporated into DNA as a purine analogue, inhibiting cell replication. Patients treated with 6‐TGua are more prone to developing skin cancer due to the photoexcitation of 6‐TGua by UVA radiation (as illustrated in the Jablonski diagram). Upon exposure to UVA, the excited 6‐TGua generates 1O2.

Thiopurines in the Management of Crohn’s Disease: Safety and Efficacy Profile in Patients with Normal TPMT Activity—A Retrospective Study

open access: yesCanadian Journal of Gastroenterology and Hepatology, 2016
Background and Aims. Thiopurines are used in the treatment of Crohn’s disease (CD) and thiopurine S-methyltransferase (TPMT) activity can guide thiopurine dosing to avoid adverse events.

Cancer Biomarker Test Utilization and Reimbursement in Australia: A National Analysis of Medicare Data

open access: yesClinical and Translational Science, Volume 19, Issue 5, May 2026.
ABSTRACT Biomarker testing plays a critical role in precision oncology by guiding targeted therapy selection. In Australia, public reimbursement for somatic tumor biomarker tests has expanded over the past decade, yet national data on utilization patterns, equity of access, and public expenditure remain limited. We identified all cancer biomarker tests

Thiopurine S-methyltransferase pharmacogenetics: insights, challenges and future directions [PDF]

open access: yesOncogene, 2006
The thiopurine S-methyltransferase (TPMT) genetic polymorphism is one of the most 'mature' examples in pharmacogenetics. That is true because of its importance clinically for the individualization of thiopurine drug therapy and also because TPMT has provided novel insights into molecular mechanisms responsible for the functional effects of common ...
