LDL and HDL transfer rates across peripheral microvascular endothelium agree with those predicted for passive ultrafiltration in humans

open access: yesJournal of Lipid Research, 2015
The mechanisms by which LDLs and HDLs cross the vascular endothelium from plasma into interstitial fluid are not understood, and have never been studied in humans in vivo.

EphB1‐Mediated Transient Blood‐Brain Barrier Opening Facilitates a Ferritin‐Based Nanotherapeutic for Alzheimer's Disease

open access: yesAdvanced Science, EarlyView.
We constructed a brain‐targeted AFn‐DPZ nanodrug delivery system and identified EphB1 as its receptor on the blood‐brain barrier (BBB). This system facilitates drug transport across the BBB and thereby ameliorates cognitive impairment in Alzheimer's disease.

Neuron‐Targeted Exosomal Delivery of siRNA Against RIPK3 Slows Neurodegenerative Progression in Alzheimer's Disease

open access: yesAdvanced Science, EarlyView.
Engineered exosomal siRNA delivery platform enables systemic, neuron‐targeted RNA transport across the blood–brain barrier. Surface functionalization with rabies virus glycoprotein‐derived peptide facilitates receptor‐mediated transcytosis, achieving efficient cytosolic delivery and robust gene silencing. Targeting RIPK3 suppresses necroptosis, reduces

Caveolin1 Is Required for Th1 Cell Infiltration, but Not Tight Junction Remodeling, at the Blood-Brain Barrier in Autoimmune Neuroinflammation

open access: yesCell Reports, 2017
Lymphocytes cross vascular boundaries via either disrupted tight junctions (TJs) or caveolae to induce tissue inflammation. In the CNS, Th17 lymphocytes cross the blood-brain barrier (BBB) before Th1 cells; yet this differential crossing is poorly ...

Sonoselective Transfection of Glioma Endothelium

open access: yesAdvanced Science, EarlyView.
Delivering non‐viral, gene‐bearing nanoparticles with thiol‐decorated surfaces to gliomas with focused ultrasound yields transfection tropism for endothelial cells. From this platform, “sonoselective” transfection of glioma endothelium is achieved via incorporation of an endothelial cell‐specific promoter.

Bone-derived PDGF-BB enhances hippocampal non-specific transcytosis through microglia-endothelial crosstalk in HFD-induced metabolic syndrome

open access: yesJournal of Neuroinflammation
Background It is well known that high-fat diet (HFD)-induced metabolic syndrome plays a crucial role in cognitive decline and brain-blood barrier (BBB) breakdown.

Strategies to Screen and Evaluate Brain Targeting Antibodies Using an iPSC-Derived Blood–Brain Barrier Model

open access: yesAntibodies
Background: Antibodies that cross the blood–brain barrier (BBB) by targeting receptor-mediated transport (RMT) systems can allow efficient drug delivery to the central nervous system (CNS).

Intranasal Delivery of Gallium–Quercetin Nanoparticles for Multi‐Target Ferroptosis Inhibition in Parkinson's Disease

open access: yesAdvanced Science, EarlyView.
Leveraging the chemical similarity between Ga3+ and Fe3+ as well as the antioxidant properties of quercetin, gallium–quercetin nanoparticles (GQNPs) were prepared to integrate iron homeostasis regulation, oxidative stress suppression, and mitochondrial protection for multi‐target ferroptosis inhibition in Parkinson's Disease.

Targeting blood-brain-barrier transcytosis – perspectives for drug delivery [PDF]

open access: yes, 2016
Efficient transcytosis across the blood-brain-barrier (BBB) is an important strategy for accessing drug targets within the central nervous system (CNS).

Transcytosis of epidermal growth factor. The epidermal growth factor receptor mediates uptake but not transcytosis

open access: yesJournal of Biological Chemistry, 1991
Madin-Darby canine kidney (MDCK) cells polarize and generate distinct apical and basolateral membrane domains when grown on permeable filter supports. Under these conditions, they transcytose fluid-phase markers. Recently, receptor-mediated transcytosis of epidermal growth factor (EGF) across MDCK cells has been reported (Maratos-Flier, E., Kao, C.-Y ...
