Role of TRPML and Two-Pore Channels in Endolysosomal Cation Homeostasis
The Journal of Pharmacology and Experimental Therapeutics, 2012The transient receptor potential (TRP) channels TRPML1, TRPML2, and TRPML3 (also called mucolipins 1-3 or MCOLN1-3) are nonselective cation channels. Mutations in the Trpml1 gene cause mucolipidosis type IV in humans with clinical features including psychomotor retardation, corneal clouding, and retinal degeneration, whereas mutations in the Trpml3 ...

MCOLN/TRPML channels in the regulation of MTORC1 and autophagy
AutophagyMCOLN1 and MCOLN3 are two Ca2+ release channels residing in the endolysosomal membrane. They are activated by phosphatidylinositol (PtdIns)-3-phosphate (PtdIns3P) and/or PtdIns(3,5)P2. Their activities are also regulated by lumenal pH, with low pH enhancing that of MCOLN1 and high pH increasing that of MCOLN3.

Transient Receptor Potential Mucolipin 1 (TRPML1) channel protein, one of the TRPML family members, is a Ca2+-permeable membrane channel with a predominant location on the late endo/lysosomes. In arterial myocytes, it has been found that this ligand-gated channel is sensitive to NAADP (Nicotinic Acid Adenine Dinucleotide Phosphate), a secondary Ca2 ...

Cellular Signalling
Autophagy is essential for eliminating aging and organelle damage that maintaining cellular homeostasis. However, the dysfunction of autophagy has been proven in hair loss such as AGA. Despite the crucial role of TRPML channels in regulating autophagy, their specific function in hair growth remains unclarified.
