DNA‐Induced Entropic Gain Triggers an Allosteric Switch for Biomolecular Condensation of Heat Shock Transcription Factor 1

open access: yesAngewandte Chemie, EarlyView.
Hsf1 undergoes phase separation around target DNA, activating stress‐responsive transcription. DNA binding tunes conformational dynamics of Hsf1DBD, which propagates through large‐scale domain rearrangements involving DBD–IDR interactions to regulate phase separation. This hierarchical framework illustrates how local structural events at the side chain

Analysis of model of tryptophan metabolism

open access: yes, 2020

Circulating Amino Acid Network Remodeling Reveals Systemic Metabolic Reprogramming Predictive of Colorectal Cancer Recurrence and Metastasis

open access: yesAdvanced Science, EarlyView.
Blood‐based amino acid patterns measured by 19F NMR reveal hidden metabolic changes in colorectal cancer. By analyzing how these amino acids interact as a network, machine learning models identify patients at higher risk of recurrence and metastasis.

Multicomponent Stapling of Glucagon‐Like Peptide‐1 Enables Receptor‐Guided PROTAC Delivery

open access: yesAngewandte Chemie, EarlyView.
We report a stapled glucagon‐like peptide‐1 (GLP‐1) analogue created via multicomponent tryptophan‐mediated Petasis reaction (TMPR). This strategy yields a stabilised peptide with superior helicity and improved potency. Conjugation to a bromodomain‐containing protein 4 (BRD4) degrader creates the first GLP‐1‐guided targeted protein degrader (PROTAC ...

Identification and validation of the important role of tryptophan-related gene CYP1B1 in the development and progression of sepsis.

open access: yesPLoS ONE
BackgroundTryptophan metabolism is involved in the progression and immune response of multiple diseases. However, the function of tryptophan metabolism in the immune characteristics of sepsis still needs to be elucidated.MethodsWe collected the datasets ...

Conformational Snapshots of CydDC in a Native Lipid Bilayer Coupling Heme Transport to Antibiotic Resistance

open access: yesAdvanced Science, EarlyView.
Phylogenetic and biochemical analyses of the heme transporter CydDC reveal its functional conservation throughout bacterial evolution and demonstrate its unique asymmetric allosteric mechanism. Furthermore, impairment of CydDC function directly affects bacterial antibiotic resistance and likely compromises antibiotic efficacy through drug efflux.
