CNS Neuroscience &Therapeutics, Volume 32, Issue 10, October 2026.We identified endoplasmic reticulum (ER) stress modulation as a convergent transcription signature across fentanyl and multiple substance use disorder (SUD)‐relevant treatments. Key ER stress genes, including TRIB3, XBP1, and DDIT3, were downregulated, while CEBPB emerged as a shared upstream regulator linked to ER stress–related proteins.

ER Stress Regulates HIV Tat and Alcohol‐Induced NLRP6 Activation in Astrocytes
CNS Neuroscience &Therapeutics, Volume 32, Issue 10, October 2026.HIV Tat and ethanol synergistically induce ER stress in astrocytes, activating an ER stress‐NLRP6 inflammasome axis that drives caspase‐1 activation and IL‐1β/IL‐18 release. Targeting ER stress or NLRP6 effectively suppresses astrocytic neuroinflammation, identifying a novel therapeutic pathway for alcohol‐exacerbated NeuroHIV.
