Identification of nurse shark V NAR single‐domain antibodies targeting the spike S2 subunit of SARS‐CoV ‐2

open access: yesFASEB Journal, 2023
Shark VNAR nanobodies targeting the S2 subunit of SARS‐CoV‐2 spike protein were identified from a naïve nurse shark VNAR phage display library by phage panning. The S2A9 VNAR showed the best neutralization activity against the original pseudotyped virus and was further characterized.

Single domain shark VNAR antibodies neutralize SARS‐CoV‐2 infection in vitro [PDF]

open access: yesFASEB Journal, 2021
Abstract Single domain shark VNAR antibodies can offer a viable alternative to conventional Ig-based monoclonal antibodies in treating COVID-19 disease during the current pandemic. Here we report the identification of neutralizing single domain VNAR antibodies selected against the SARS-CoV-2 spike protein derived from

Preparation of Immune and Synthetic VNAR Libraries as Sources of High-Affinity Binders

2022
The shark-derived autonomous variable antibody domains known as VNARs are attractive tools for therapeutic and diagnostic applications due to their favorable properties like small size (approximately 12 kDa), high thermal and chemical stability, and good tissue penetration.

Construction of Semisynthetic Shark vNAR Yeast Surface Display Antibody Libraries

2023
The adaptive immune system of sharks comprises a unique heavy chain-only antibody isotype, termed immunoglobulin new antigen receptor (IgNAR), in which antigen binding is mediated by a single variable domain, referred to as vNAR. In recent years, efforts were made to harness these domains for biomedical and biotechnological applications particularly ...

Diagnostic and therapeutic potential of shark variable new antigen receptor (VNAR) single domain antibody

open access: yesInternational Journal of Biological Macromolecules, 2020
Conventional monoclonal antibodies (mAbs) have been widely used in research and diagnostic applications due to their high affinity and specificity. However, multiple limitations, such as large size, complex structure and sensitivity to extreme ambient temperature potentially weaken the performance of mAbs in certain applications.

Shark VNAR phage display libraries: An alternative source for therapeutic and diagnostic recombinant antibody fragments

Fish & Shellfish Immunology, 2023
The development of recombinant antibody fragments as promising alternatives to full-length immunoglobulins offers vast opportunities for biomedicine. Antibody fragments have important advantages compared with conventional monoclonal antibodies that make them attractive for the biotech industry: superior stability and solubility, reduced immunogenicity,

Cytoplasmic and periplasmic expression of recombinant shark VNAR antibody in Escherichia coli

Preparative Biochemistry and Biotechnology, 2019
Shark variable new antigen receptors (VNARs) are known to possess excellent heat-stability, and the long complementarity determining region 3 (CDR3) has permitted it to penetrate into the cleft region of antigens. The number of cysteine (Cys) residues contained within VNAR is greater than in conventional antibodies, entailing disulfide bond formation ...

Generation of VNAR Libraries from Immunized Sharks and Selection of Target-Specific Clones

2021
Cartilaginous fishes (sharks, skates, rays, and chimeras) are the most phylogenetically distant lineage relative to mammals in which somatically rearranging immunoglobulins (Igs or antibodies) have also been found. Alongside their conventional (heavy-light chain) isotypes, IgM and IgW, sharks produce the novel isotype, IgNAR, a heavy-chain homodimer ...
