Synthesis and Biological Evaluation (in Vitro and in Vivo) of Cyclic RGD Peptidomimetic - Paclitaxel Conjugates Targeting Integrin alphaVbeta3 [PDF]

open access: yes, 2012
A small library of integrin ligand - Paclitaxel conjugates 10-13 was synthesized with the aim of using the tumor-homing cyclo[DKP-RGD] peptidomimetics for site-directed delivery of the cytotoxic drug.

Legumain Restrains Granuloma Formation by Inhibiting mTORC1/STAT1‐Mediated M1 Macrophage Polarization in Sarcoidosis

open access: yesAdvanced Science, EarlyView.
Legumain (LGMN) is upregulated in macrophages during sarcoid‑like granuloma formation. Macrophage‑derived LGMN binds to integrin αvβ3 and suppresses mTORC1/STAT1 signaling to restrain M1 macrophage polarization. Intratracheal delivery of lipid nanoparticles carrying Lgmn plasmid DNA (pDNA) elevates LGMN expression and effectively attenuates pulmonary ...

Selective Inhibition of Integrin β3 Topology Provides a Safer Antithrombotic Strategy

open access: yesAdvanced Science, EarlyView.
Current integrin αIIbβ3 inhibitors effectively reduce thrombosis but also increase bleeding risk. During thrombosis, high shear blood flow can directly activate the integrin αIIbβ3 via a distinct topological change in the β3 transmembrane domain, independent of hemostatic platelet signaling.

Sono‐Mechanogenetics: Linking Ultrasound Physics With Cellular Mechanobiology

open access: yesAdvanced Science, EarlyView.
Sono‐mechanogenetics links ultrasound physics with cellular mechanotransduction to enable noninvasive control of engineered biological systems. Acoustic forces generate distinct deformation modes that activate intracellular signaling pathways, which can be coupled to synthetic gene circuits to regulate diverse cellular functions, including gene ...

Brag2 differentially regulates β1- and β3-integrin-dependent adhesion in endothelial cells and is involved in developmental and pathological angiogenesis [PDF]

open access: yes, 2016
β1-Integrins are essential for angiogenesis. The mechanisms regulating integrin function in endothelial cells (EC) and their contribution to angiogenesis remain elusive.

Mechanically Triggered DNA Nanovehicles for Targeted Dual‐Drug Cancer Therapy

open access: yesAdvanced Science, EarlyView.
A mechanically triggered DNA nanovehicle is introduced that co‐releases two anticancer drugs in response to integrin‐mediated cellular forces. Cholesterol‐anchored DNA assemblies undergo force‐induced unfolding at cell–cell junctions, enabling selective, localized dual‐drug activation in cancer cells while minimizing off‐target toxicity in low‐tension ...

Pharmacological and Clinical Importance of Integrin Antagonists in Treatment of Cancer [PDF]

open access: yes, 2015
Integrins are heterodimeric molecules that are composed of 18 α -subunits and 8 β -subunits. They exist in 24 distinctive shapes based on combination of these sub-units and are mainly responsible for the adhesion of extracellular ...

Dual Physiological Barriers Bypassed by a Silk‐Based Supramolecular Protein Delivery Platform for Neuroinflammation Mitigation in Alzheimer's Disease

open access: yesAdvanced Science, EarlyView.
A supramolecular nanocomplex based on phenolic‐modified silk sericin is developed to deliver antioxidant enzymes across the blood–brain barrier and escape lysosomal degradation. This dual‐barrier‐crossing system reduces neuroinflammation and improves cognitive performance in Alzheimer's disease mouse model, offering a promising strategy for protein ...

Bimodal anti-glioma mechanisms of cilengitide demonstrated by novel invasive glioma models [PDF]

open access: yes, 2013
Integrins are expressed in tumor cells and tumor endothelial cells, and likely play important roles in glioma angiogenesis and invasion. We investigated the anti-glioma mechanisms of cilengitide (EMD121974), an v3 integrin inhibitor, utilizing the novel ...

The HCMV‐encoded miR‐UL36‐3p promotes angiogenesis of endothelial cells by downregulating FOXO3

open access: yesAnimal Models and Experimental Medicine, EarlyView.
This illustration shows the role of hcmv‐miR‐UL36‐3p in endothelial cell angiogenesis. During productive infection, endothelial cells express HCMV‐encoded miRNAs. Viral hcmv‐miR‐UL36‐3p promotes endothelial cell proliferation, migration, and capillary tube formation by directly targeting and downregulating FOXO3. The pro‐angiogenic activity of hcmv‐miR‐
