Novel Approach for CES1 Genotyping: Integrating Single Nucleotide Variants and Structural Variation [PDF]

open access: yesPharmacogenomics, 2018
Development of a specific procedure for genotyping of CES1A1 (CES1) and CES1A2, a hybrid of CES1A1  and the pseudogene CES1P1.The number of CES1A1 and CES1A2  copies and that of CES1P1  were determined using real-time PCR. Long range PCRs followed by secondary PCRs allowed sequencing of single nucleotide variants in CES1A1 and CES1A2.A procedure ...

Bioactive lipid metabolism by carboxylesterase 1 (CES1) in macrophages (LB154)

open access: yesThe FASEB Journal, 2014
Human carboxylesterases (CES) have an emerging role in lipid metabolism. CES1 in macrophages is a hydrolase with broad endogenous substrate specificity, including cholesteryl ester, triacylglycerol, and 2‐arachidonoylglycerol (2‐AG), which may be relevant to cardiovascular disease and other pathologies related to lipid metabolism dysfunction.

Analysis of a gene panel for targeted sequencing of colorectal cancer samples [PDF]

open access: yes, 2018
Colorectal cancer (CRC) is a leading cause of death worldwide. Surgical intervention is a successful treatment for stage I patients, whereas other more advanced cases may require adjuvant chemotherapy.

Molecular Profiling of Genes Associated With Methylphenidate Pathway Therapy and Discovery of New Variants in Amazonian Amerindian Populations

open access: yesAmerican Journal of Medical Genetics Part B: Neuropsychiatric Genetics, EarlyView.
ABSTRACT In Attention Deficit Hyperactivity Disorder (ADHD), methylphenidate is one of the most widely used drugs, in which patient response significantly impacts prognosis. This study aimed to characterize the molecular profile of 10 genes associated with methylphenidate therapy.

A BET Protein Inhibitor Targeting Mononuclear Myeloid Cells Affects Specific Inflammatory Mediators and Pathways in Crohn’s Disease

open access: yesCells, 2022
Background: Myeloid cells are critical determinants of the sustained inflammation in Crohn’s Disease (CD). Targeting such cells may be an effective therapeutic approach for refractory CD patients. Bromodomain and extra-terminal domain protein inhibitors (

Genetic Analysis Reveals a Significant Contribution of CES1 to Prostate Cancer Progression in Taiwanese Men [PDF]

open access: yesCancers, 2020
The genes that influence prostate cancer progression remain largely unknown. Since the carboxylesterase gene family plays a crucial role in xenobiotic metabolism and lipid/cholesterol homeostasis, we hypothesize that genetic variants in carboxylesterase genes may influence clinical outcomes for prostate cancer patients. A total of 478 (36 genotyped and

Quantitative Analysis Reveals Hitchhiking Drives Polysorbate Hydrolase Persistence Via Host Cell Protein–Antibody Interactions

open access: yesBiotechnology and Bioengineering, EarlyView.
ABSTRACT Polysorbate‐degrading host cell proteins (HCPs) represent a critical challenge in the manufacturing of monoclonal antibody therapeutics due to their potential to persist during downstream processing. While their enzymatic activity has been characterized, the role of direct HCP‐mAb interactions, particularly those involving polysorbate ...

ProTides for Antiviral Activity Beyond Liver Cells

open access: yesChemistry – A European Journal, EarlyView.
A strategy for obtaining prodrugs of antiviral nucleotides with broad tissue activity is presented that relies on cycloalkyl or cycloalkylalkyl esters, improving uptake and esterase cleavage, and producing nanomolar inhibitors in kidney, colon, and lung cells.

Hypolipidemic agent Z-guggulsterone: metabolism interplays with induction of carboxylesterase and bile salt export pump

open access: yesJournal of Lipid Research, 2012
Z-Guggulsterone is a major ingredient in the Indian traditional hypolipidemic remedy guggul. A study in mice has established that its hypolipidemic effect involves the farnesoid X receptor (FXR), presumably by acting as an antagonist of this receptor. It

Pharmacogenetics of Carboxylesterase 1 [PDF]

open access: yes, 2017
Carboxylesterases (CESs) catalyze the hydrolysis of a variety of ester- and amide containing compounds to their respective free acids. The main CES isozymes involved in drug metabolism are carboxylesterases 1 and 2 (CES1 and CES2). CES1 contributes to an
