Development of Biolayer Interferometry (BLI)-Based Double-Stranded RNA Detection Method with Application in mRNA-Based Therapeutics and Vaccines

open access: yesPharmaceutics
Background: In vitro-transcribed (IVT) mRNA has been established as a promising platform for therapeutics and vaccine development. Double-stranded RNA (dsRNA) is a major impurity of IVT mRNA and can trigger unfavored immune responses, potentially causing

Allosteric Regulation of RNA Affinity by Motif V‐VI Coupling in West Nile Virus NS3 Helicase

open access: yesProteins: Structure, Function, and Bioinformatics, EarlyView.
ABSTRACT The rise of flaviviral diseases, including West Nile virus (WNV), presents a growing threat to global public health and underscores the urgent need for new therapeutic strategies. The non‐structural protein 3 helicase (NS3h) of the Orthoflavivirus genus, including WNV, is essential for viral replication and a promising antiviral target ...

RNA degradomics and proteomics reveal the mechanism of dsProsβ1‐mediated proteasome targeting in Psylliodes chrysocephala

open access: yesPest Management Science, EarlyView.
RNA degradomics revealed dsProsβ1‐derived siRNA‐mediated mRNA cleavage events, mainly at uracil‐guanine and adenine‐adenine pairs. Proteasome inhibition via dsProsβ1 increased mitochondrial and cytoskeletal proteins while reducing translation‐related and mRNA‐binding proteins.

Reporters transiently transfected into mammalian cells are highly sensitive to translational repression induced by dsRNA expression.

open access: yesPLoS ONE, 2014
In mammals, double-stranded RNA (dsRNA) can mediate sequence-specific RNA interference, activate sequence-independent interferon response, or undergo RNA editing by adenosine deaminases. We showed that long hairpin dsRNA expression had negligible effects

Assessment of RNAi-induced silencing in banana (Musa spp.) [PDF]

open access: yes, 2015
In plants, RNA- based gene silencing mediated by small RNAs functions at the transcriptional or post-transcriptional level to negatively regulate target genes, repetitive sequences, viral RNAs and/or transposon elements.

Synthetic polymer nanocarrier enhances dsRNA stability but not RNAi efficacy in Brassicogethes aeneus

open access: yesPest Management Science, EarlyView.
Experimental setup for dsRNA feeding in Brassicogethes aeneus to evaluate the effect of a synthetic polymer nanocarrier on RNAi efficiency. The experiment assessed whether the nanocarrier enhances RNAi after oral dsRNA delivery. (a) Stability of dsRNA–nanocarrier complexes was tested in simulated gut conditions. (b) Gene expression of αCOP and dre4 was

Exogenous dsRNA triggers sequence-specific RNAi and fungal stress responses to control Magnaporthe oryzae in Brachypodium distachyon

open access: yesCommunications Biology
In vertebrates and plants, dsRNA plays crucial roles as PAMP and as a mediator of RNAi. How higher fungi respond to dsRNA is not known. We demonstrate that Magnaporthe oryzae (Mo), a globally significant crop pathogen, internalizes dsRNA across a broad ...

Utilization of Bacteriophage phi6 for the Production of High-Quality Double-Stranded RNA Molecules

open access: yesViruses
Double-stranded RNA (dsRNA) molecules are mediators of RNA interference (RNAi) in eukaryotic cells. RNAi is a conserved mechanism of post-transcriptional silencing of genes cognate to the sequences of the applied dsRNA.

The NS3 protein of rice hoja blanca virus suppresses RNA silencing in mammalian cells [PDF]

open access: yes, 2008
The NS3 protein of the tenuivirus rice hoja blanca virus (RHBV) has previously been shown to represent the viral RNA interference (RNAi) suppressor and is active in both plant and insect cells by binding short interfering RNAs (siRNAs) in vitro.

Gene‐specific double‐stranded RNAs induce mortality in the South African mealybug Delottococcus aberiae

open access: yesPest Management Science, EarlyView.
Gene‐specific double‐stranded RNAs effectively silence essential genes in the invasive citrus pest Delottococcus aberiae, leading to significant mortality through both microinjection and oral delivery. These results provide proof of concept for RNA interference‐based biopesticides as selective and sustainable tools for citrus integrated pest management.
