Insulin Resistance: An Update on Biochemical and Pathophysiological Mechanisms and Impact on Various Diseases

open access: yesiNew Medicine, EarlyView.
ABSTRACT Insulin resistance is the biological phenomenon in which the human body's normal response to the metabolic hormone insulin is compromised. Insulin is a regulator of most of the essential metabolic steps in the body that control energy homoeostasis, so dysregulation leads to multiple diverse human diseases including, most prominently, Type 2 ...

MAIT Cells Suppress IgE‐Mediated Asthma via IFNγ‐Dependent B Cell Regulation

open access: yesAllergy, EarlyView.
In this study, we demonstrate that MAIT cell antagonism during sensitization and challenge with HDM worsens the development of airway hyperreactivity without effects on type 2 or 17 associated cytokine production or lung inflammation. Rather, MAIT cells appear to regulate HDM‐induced asthma through direct inhibition of IgE production.

CD169(+) macrophages present lipid antigens to mediate early activation of iNKT cells in lymph nodes

open access: yes, 2010
Invariant natural killer T cells (iNKT cells) are involved in the host defense against microbial infection. Although it is known that iNKT cells recognize glycolipids presented by CD1d, how and where they encounter antigen in vivo remains unclear.

Specific deletion of LDL receptor-related protein on macrophages has skewed in vivo effects on cytokine production by invariant natural killer T cells. [PDF]

open access: yesPLoS ONE, 2014
Expression of molecules involved in lipid homeostasis such as the low density lipoprotein receptor (LDLr) on antigen presenting cells (APCs) has been shown to enhance invariant natural killer T (iNKT) cell function. However, the contribution to iNKT cell

4‐1BB Expression and Signaling Regulates MAIT Cell Activation and Effector Functions

open access: yesImmunology &Cell Biology, EarlyView.
4‐1BB is a highly expressed TNFRSF co‐stimulatory receptor on MAIT cells upon activation. Expression and signaling via 4‐1BB enhance activation and provide co‐stimulation, boosting MAIT cell effector responses to TCR‐mediated activation. ABSTRACT Mucosal associated invariant T (MAIT) cells are abundant unconventional T cells that recognize microbe ...

Innovations in Obesity Treatment: Beyond Adipose Tissue Dysfunction

open access: yesObesity Reviews, EarlyView.
Obesity drives chronic inflammation, insulin resistance, type 2 diabetes, and cancer development through adipocyte dysfunction. Addressing this multisystemic disorder requires integrated strategies beyond diet and exercise, such as thermogenesis activation via menthol or capsinoids and appetite control through GLP‐1/GIP agonists and neuromodulation to ...

Professional Differences in Antigen Presentation to iNKT Cells [PDF]

open access: yesImmunity, 2014
Invariant natural killer T cells are preactivated lymphocytes that react upon recognition of CD1d-antigen complexes. Accordingly, any type of CD1d-positive cell could behave as antigen-presenting cell (APC). In this issue of Immunity, Arora et al. (2014), report that professional APCs still make the difference.

Development and function of invariant natural killer T cells producing T(h)2- and T(h)17-cytokines. [PDF]

open access: yesPLoS Biology, 2012
There is heterogeneity in invariant natural killer T (iNKT) cells based on the expression of CD4 and the IL-17 receptor B (IL-17RB), a receptor for IL-25 which is a key factor in T(H)2 immunity.

A party of three: iNKT cells in GVHD prevention [PDF]

open access: yesBlood, 2015
In this issue of Blood, Schneidawind et al demonstrate that the adoptive transfer of CD4+ invariant natural killer T (iNKT) cells from third-party mice protects from lethal graft-versus-host disease (GVHD) through expansion of donor regulatory T cells (Tregs) in a murine model of allogeneic hematopoietic cell transplantation (HCT).

Motility and tumor infiltration are key aspects of invariant natural killer T cell anti-tumor function

open access: yesNature Communications
Dysfunction of invariant natural killer T (iNKT) cells contributes to immune resistance of tumors. Most mechanistic studies focus on their static functional status before or after activation, not considering motility as an important characteristic for ...
