DOR/Tp53inp2 and Tp53inp1 constitute a metazoan gene family encoding dual regulators of autophagy and transcription. [PDF]

open access: yesPLoS ONE, 2012
Human DOR/TP53INP2 displays a unique bifunctional role as a modulator of autophagy and gene transcription. However, the domains or regions of DOR that participate in those functions have not been identified.

LDHB suppresses the PDCoV proliferation by targeting viral nucleocapsid protein for autophagic degradation

open access: yesMicrobiology Spectrum
Porcine deltacoronavirus (PDCoV) is a newly identified enteric coronavirus that causes serious diarrhea and vomiting in pigs, leading to substantial economic losses globally.

Autophagy Promotes Focal Adhesion Disassembly and Cell Motility of Metastatic Tumor Cells through the Direct Interaction of Paxillin with LC3

open access: yesCell Reports, 2016
Autophagy is a conserved catabolic process that plays a housekeeping role in eliminating protein aggregates and organelles and is activated during nutrient deprivation to generate metabolites and energy.

YIPF3 and YIPF4 regulate autophagic turnover of the Golgi apparatus

open access: yesThe EMBO Journal
The degradation of organelles by autophagy is essential for cellular homeostasis. The Golgi apparatus has recently been demonstrated to be degraded by autophagy, but little is known about how the Golgi is recognized by the forming autophagosome.

Wnt signalosome assembly is governed by conformational flexibility of Axin and by the AP2 clathrin adaptor

open access: yesNature Communications
Wnt signal transduction relies on the direct inhibition of GSK3 by phosphorylated PPPSPxS motifs within the cytoplasmic tail of the LRP6 co-receptor. How GSK3 is recruited to LRP6 remains unclear.

Structural and Functional analysis of the GABARAP interaction motif (GIM) [PDF]

open access: yes, 2017
Through the canonical LC3 interaction motif (LIR), [W/F/Y]-X1-X2-[I/L/V], protein complexes are recruited to autophagosomes to perform their functions as either autophagy adaptors or receptors.

Elucidation of the anti-autophagy mechanism of the Legionella effector RavZ using semisynthetic LC3 proteins

open access: yeseLife, 2017
Autophagy is a conserved cellular process involved in the elimination of proteins and organelles. It is also used to combat infection with pathogenic microbes.

ClustalO alignment of kinetoplastid ATG2 protein sequences surrounding the conserved ATG8-binding LIR motif (above).

open access: yes
To illustrate conservation, the Caenorhabditis elegans motif (conserved up to human ATG2A and ATG2B) is shown as a reference. The lack of perfect alignment in certain species is likely due to very low conservation of the surrounding sequence.

Hypoxic mitophagy regulates mitochondrial quality and platelet activation and determines severity of I/R heart injury

open access: yeseLife, 2016
Mitochondrial dysfunction underlies many prevalent diseases including heart disease arising from acute ischemia/reperfusion (I/R) injury. Here, we demonstrate that mitophagy, which selectively removes damaged or unwanted mitochondria, regulated ...

Differential roles of N- and C-terminal LIR motifs in the catalytic activity and membrane targeting of RavZ and ATG4B proteins

open access: yesBMB Reports
Mammalian ATG8 proteins (mATG8s) are essential for selective autophagy because they recruit various proteins with LC3- interacting region (LIR) motifs to autophagic membranes. The RavZ protein, secreted by Legionella pneumophila, and mammalian ATG4B possess functional LIR motifs that participate in lipidated mATG8 deconjugation on autophagic membranes.
