Dissection of malonyl-coenzyme A reductase of Chloroflexus aurantiacus results in enzyme activity improvement. [PDF]
The formation of fusion protein in biosynthetic pathways usually improves metabolic efficiency either channeling intermediates and/or colocalizing enzymes. In the metabolic engineering of biochemical pathways, generating unnatural protein fusions between

Properties of Malonyl‐CoA Decarboxylase and its Relation with Malonyl‐CoA Incorporation into Fatty Acids by Rat Liver Mitochondria [PDF]
1 Malonyl-CoA decarboxylase in rat liver, heart, kidney and brain is mainly a mitochondrial enzyme associated with the matrix space. 2 The Michaelis constant for this enzyme, in rat liver mitochondria, has been calculated as 42 μM. 3 N-Ethylmaleimide, p-chloromercuribenzoate and palmityl-CoA, but not arsenite, inhibit malonyl-CoA decarboxylase.

Mechanism of activation of bovine kidney pyruvate dehydrogenase a kinase by malonyl‐CoA and enzyme‐catalyzed decarboxylation of malonyl‐CoA [PDF]
The activity of the pyruvate dhydrogenase kinase, which phosphorylates and therby inactivates the pyruvate dehydrogenase complex, was stimulated by malonyl‐CoA. Treatment with [2‐14C]malonyl‐CoA resulted in acylation of sites in the complex. Both acylation and activation of kinase activity increased in a time‐dependent manner with a parallel increase ...

A Genetically Encoded Metabolite Sensor for Malonyl-CoA
Malonyl-CoA is the rate-determining metabolite for long chain de novo fatty acid synthesis and allosterically inhibits the rate-setting step in long chain fatty acid β-oxidation. We developed a cell-based genetically encoded biosensor based on the malonyl-CoA responsive Bacillus subtilis transcriptional repressor, FapR, for living mammalian cells. Here,

A stereoselective functionalization method of cyclic imines under mild organocatalytic conditions was developed. Bifunctional cinchona alkaloid catalysts combined with carefully optimized reaction conditions to suppress background reactivity enabled highly enantio‐ and diastereoselective addition reactions of malonates to cyclic imines.

De novo synthesis and elongation of fatty acids by subcellular fractions of monkey aorta
Subcellular fractions of aorta of squirrel monkey (Saimiri sciureus) were examined for their ability to synthesize and elongate fatty acids. High-speed supernate (HSS) incorporated substantial quantities of malonyl CoA into fatty acids while acetyl CoA ...

Production of 3-hydroxypropionic acid (3-HP) in Pichia pastoris (syn. Komagataella phaffii) via the malonyl-CoA pathway has been recently demonstrated using glycerol as a carbon source, but the reported metrics were not commercially relevant.

Sensing of nutrients by CPT1C regulates late endosome/lysosome anterograde transport and axon growth
Anterograde transport of late endosomes or lysosomes (LE/Lys) is crucial for proper axon growth. However, the role of energetic nutrients has been poorly explored.

Ralstonia solanacearum RSp0194 Encodes a Novel 3-Keto-Acyl Carrier Protein Synthase III. [PDF]
Fatty acid synthesis (FAS), a primary metabolic pathway, is essential for survival of bacteria. Ralstonia solanacearum, a β-proteobacteria member, causes a bacterial wilt affecting more than 200 plant species, including many economically important plants.
