The FOXC2‐LAMA4 Axis Orchestrates Vasculogenic Mimicry and Immunosuppressive Niche Formation to Drive Metastatic Cascade in Renal Cell Carcinoma

Advanced clear cell renal cell carcinoma metastasis is driven by a FOXC2+ tumor subpopulation. These cells execute vasculogenic mimicry for dissemination and secrete LAMA4. LAMA4 engages macrophage ITGA6, activating the STAT6/GATA3 axis to polarize macrophages into a pro‐metastatic, immunosuppressive phenotype.

SLC2A3‐Mediated Lactate Metabolism Promotes Lung Cancer Bone Metastasis by Modulating P53 Lactylation and Immune Evasion

SLC2A3 derived lactate promotes metastasis through p53 lactylation at K120 and osteoclast differentiation. Pharmacological inhibition of SLC2A3 upregulates PD‐1 expression on CD8+ T cells via lactate induced p53 lactylation, which modulates immune evasion. Human islet amyloid polypeptide (hIAPP) was used to mimic T2DM, and Aβ42‐hIAPP co‐oligomers were delivered into the human mature cerebral organoids (COs), which reproduce typical AD pathology and significant neuronal death more closely resembling that of AD patients.
