Abstract Approximately half of all cases of Infantile Epileptic Spasms Syndrome (IESS) do not respond to vigabatrin and hormonal therapies. There is no clear consensus as to the second‐line therapy for IESS. Ketogenic diet (KD) has emerged as an effective treatment for certain drug‐resistant epilepsies and in many cases of IESS.

Tumour cells are known to be highly glycolytic, thus producing high amounts of lactic acid. Monocarboxylate transporters (MCTs), by promoting the efflux of the accumulating acids, constitute one of the most important mechanisms in the maintenance of tumour intracellular pH.

This review highlights recent advances in mitochondria‐targeted nanomedicine for cancer therapy. It summarizes key targeting strategies and delineates four principal therapeutic mechanisms: oxidative stress–induced mitochondrial dysfunction, disruption of energy metabolism, direct mitochondrial damage, and induction of immunogenic cell death.
