Effects of hypo O-GlcNAcylation on Drosophila development [PDF]

open access: yes, 2018
Post-translational modification of serine/threonine residues in nucleocytoplasmic proteins with N-acetylglucosamine (OGlcNAcylation) is an essential regulatory mechanism in many cellular processes.

Key Regulators of Parasite Biology Viewed Through a Post‐Translational Modification Repertoire

open access: yesPROTEOMICS, Volume 25, Issue 21-22, Page 253-276, November 2025.
ABSTRACT Parasites are the leading causes of morbidity and mortality in both humans and animals, imposing substantial socioeconomic burdens worldwide. Controlling parasitic diseases has become one of the key issues in achieving “One Health”. Most parasites have sophisticated life cycles exhibiting progressive developmental stages, morphologies, and ...

Glucose and glutamine fuel protein O-GlcNAcylation to control T cell self-renewal and malignancy [PDF]

open access: yes, 2016
Sustained glucose and glutamine transport are essential for activated T lymphocytes to support ATP and macromolecule biosynthesis. We now show that glutamine and glucose also fuel an indispensible dynamic regulation of intracellular protein O ...

Decoding Glycosylation in Neurodegenerative Diseases: Mechanistic Insights and Therapeutic Opportunities

open access: yesThe FASEB Journal, Volume 39, Issue 20, 31 October 2025.
Graphical summary of glycosylation impairments in various neurodegenerative disorders. Among the diverse forms of glycosylation, the most extensively studied include N‐glycosylation, O‐glycosylation, and O‐GlcNAcylation. Aberrant glycosylation has been implicated in multiple neurodegenerative conditions.

Liver X receptor regulates hepatic nuclear O-GlcNAc signaling and carbohydrate responsive element-binding protein activity [PDF]

open access: yes, 2015
Liver X receptor (LXR)α and LXRβ play key roles in hepatic de novo lipogenesis through their regulation of lipogenic genes, including sterol regulatory element-binding protein (SREBP)-1c and carbohydrate responsive element-binding protein (ChREBP).

A Genetically Encoded Assay System to Quantify O‐GlcNAc Transferase (OGT) Activity in Live Cells

open access: yesAngewandte Chemie, Volume 137, Issue 43, October 20, 2025.
A genetically encoded, imaging‐based reporter enables direct measurement of O‐GlcNAc transferase (OGT) activity in living cells. The system exploits OGT‐dependent proteolysis of HCF‐1 to drive nuclear and cytoplasmic redistribution of fluorescent signals, allowing cell‐by‐cell spatially resolved quantification of OGT activity.

The role of O-GlcNAcase during heart failure.

open access: yes, 2016
Global augmentation of protein O-GlcNAcylation occurs in response to a myriad of stressors and confers a survival advantage at the cellular level. This protective phenomenon has been demonstrated to mediate cardioprotection through various in vitro and in vivo studies during ischemia-reperfusion, myocardial infarction, and oxidative stress; however ...

O-GlcNAc impacts mitophagy via the PINK1-dependent pathway

open access: yesFrontiers in Aging Neuroscience
BackgroundThe accumulation of dysfunctional mitochondria is an early feature of Alzheimer’s disease (AD). The impaired turnover of damaged mitochondria increases reactive oxygen species production and lowers ATP generation, leading to cellular toxicity ...

The Many Ways by Which O-GlcNAcylation May Orchestrate the Diversity of Complex Glycosylations

open access: yesMolecules, 2018
Unlike complex glycosylations, O-GlcNAcylation consists of the addition of a single N-acetylglucosamine unit to serine and threonine residues of target proteins, and is confined within the nucleocytoplasmic and mitochondrial compartments. Nevertheless, a

Elevation of nucleocytoplasmic β-N-acetylglucosaminidase (O-GlcNAcase) activity in thyroid cancers

open access: yesInternational Journal of Molecular Medicine, 2010
Single N-acetylglucosamine residues attached by O-linkage to serine or threonine (O-GlcNAc) are an abundant, dynamic and inducible post-translational modification of cytoplasmic and nuclear proteins. This study analyzes the activity of the enzyme involved in the removal of these sugar residues, i.e. beta-N-acetylglucosaminidase (O-GlcNAcase) as well as
