Polyketide synthesis in vitro on a modular polyketide synthase [PDF]

open access: yesChemistry & Biology, 1995
The 6-deoxyerythronolide B synthase (DEBS) of Saccharopolyspora erythraea, which synthesizes the aglycone core of the antibiotic erythromycin A, contains some 30 active sites distributed between three multienzyme polypeptides (designated DEBS1-3). This complexity has hitherto frustrated mechanistic analysis of such enzymes.

Quantification of N-acetylcysteamine activated methylmalonate incorporation into polyketide biosynthesis [PDF]

open access: yesBeilstein Journal of Organic Chemistry, 2013
Polyketides are biosynthesized through consecutive decarboxylative Claisen condensations between a carboxylic acid and differently substituted malonic acid thioesters, both tethered to the giant polyketide synthase enzymes.

First Identification of Amphidinols from Mexican Strains and New Analogs

open access: yesToxins, 2023
The genus Amphidinium has been the subject of recent attention due to the production of polyketide metabolites. Some of these compounds have shown significant bioactivities and could be related to species interactions in the natural benthic ...

Biosynthesis of Fungal Natural Products Involving Two Separate Pathway Crosstalk

open access: yesJournal of Fungi, 2022
Fungal natural products (NPs) usually possess complicated structures, exhibit satisfactory bioactivities, and are an outstanding source of drug leads, such as the cholesterol-lowering drug lovastatin and the immunosuppressive drug mycophenolic acid.
