Coupling of both a transactivation module and a double‐stranded DNA‐binding domain boosts Cas12i3 variant‐based cytosine and adenine editing in plants

open access: yesJournal of Integrative Plant Biology, Volume 68, Issue 9, Page 3196-3207, September 2026.
Fusions of a transactivation module and a double‐stranded DNA‐binding domain produced effective Cas12i3 variant‐based cytosine and adenine base editors, with improved C‐to‐T and A‐to‐G base editing efficiency and expanded editing window. Generated herbicide‐resistant rice germplasm by using these base editors demonstrates their potential for precise ...

Additional file 1: of Protoplast isolation, transient transformation of leaf mesophyll protoplasts and improved Agrobacterium-mediated leaf disc infiltration of Phaseolus vulgaris: tools for rapid gene expression analysis

open access: yes, 2016
Selection of appropriate Phaseolus vulgaris tissue material for protoplast isolation. (A) Wing and keel petals were excised, retaining the pink-colored portions, for protoplast isolation. (B) The roots of 3-day-old germinated seeds were cut 3 mm from tip

Enhanced exonuclease–Cas9 systems promote multiple nucleotide deletions with higher efficiency and broader targeting scope in plants

open access: yesJournal of Integrative Plant Biology, Volume 68, Issue 9, Page 3316-3328, September 2026.
Cas9 preferentially generates small indels. By tethering an exonuclease and a DNA‐binding domain (DBD) to Cas9, the Exo‐DBD‐Cas9 system efficiently promotes multiple nucleotide deletions. These expanded deletions enable effective disruption of miRNA‐mediated regulation and relieve translational repression through targeted editing of 3′ untranslated ...

Synergistic engineering of Casδ nuclease for robust genome editing

open access: yesJournal of Integrative Plant Biology, Volume 68, Issue 9, Page 3231-3242, September 2026.
A hierarchical engineering strategy was used to enhance the editing activity of Casδ, yielding an optimized variant termed enCasδ. This variant enables robust genome editing in animal cells and plants, with overall editing performance comparable to that of SpCas9 and other Cas12 nucleases.

SIB1‐SEC23A undergo ER to chloroplast relocalization to mediate immunity in Arabidopsis thaliana

open access: yesJournal of Integrative Plant Biology, Volume 68, Issue 9, Page 3458-3472, September 2026.
SEC23A mediates membrane trafficking between the endoplasmic reticulum (ER) and Golgi apparatus and interacts with the immunity protein SIGMA FACTOR‐BINDING PROTEIN1 in Arabidopsis. Under stress, both proteins relocate from the ER to chloroplasts, suppressing photosynthesis genes and enhancing defense genes.

Engineering glyphosate‐resistant watermelon by prime editing

open access: yesJournal of Integrative Plant Biology, Volume 68, Issue 9, Page 3298-3300, September 2026.
Editing the watermelon EPSPS gene using a prime editing platform with visible markers created a non‐transgenic glyphosate‐resistant line. These robust heterozygous mutants tolerate field‐level herbicide doses without growth penalties, serving as ideal parental lines for crop breeding.

The ER–Golgi Lipid Exchanger PsORP1 Is Essential for Phytophthora sojae Development and Is Blocked by Oxathiapiprolin via Its OSBP‐Related Domain (ORD)

open access: yesMolecular Plant Pathology, Volume 27, Issue 9, September 2026.
PsORP1 anchors ER–Golgi MCSs to mediate PS/PI4P counter‐transport, and oxathiapiprolin targets the ORD to inhibit this lipid exchange. ABSTRACT Oxysterol‐binding protein (OSBP)‐related proteins (ORPs) are indispensable eukaryotic lipid transporters and promising targets for pharmaceutical and agrochemical exploitation.

The Conserved Guanyl‐Specific Ribonuclease T1 Effector EC19 Is Required for Full Virulence of the Vascular Wilt Fungus Fusarium oxysporum on Tomato

open access: yesMolecular Plant Pathology, Volume 27, Issue 9, September 2026.
EC19, a secreted ribonuclease T1 effector from Fusarium oxysporum f. sp. lycopersici (Fol), is required for tomato wilt virulence. EC19‐overexpressing tomato lines display increased susceptibility to Fol and altered transcriptional changes associated with plant hormone signal transduction, plant–pathogen interaction, and phenylpropanoid biosynthesis ...

Systematic Identification and Functional Characterisation of Colletotrichum fructicola Effectors During Camellia oleifera Colonisation

open access: yesMolecular Plant Pathology, Volume 27, Issue 9, September 2026.
Five novel effectors were identified in Colletotrichum fructicola required for full virulence. They suppress host immunity, modulate antioxidant enzymes and reprogramme metabolism in Camellia oleifera, providing targets for anthracnose control. ABSTRACT Camellia oleifera is an important woody oil crop in southern China, but its production is severely ...
