Alternative splicing (AS) expands proteomic diversity and functional complexity in eukaryotes, regulated by spliceosomal components, RNA elements, and epigenetic modifications. Dysregulated AS contributes to diseases, including cancer, neurodegenerative disorders, and cardiovascular conditions, among others. Therapeutic interventions, such as antisense

Mining Alzheimer's Interactomes, Macromolecular Complexes and Pathways for Drug Discovery
ABSTRACT Alzheimer's disease (AD) is a progressive neurodegenerative disorder that leads to dementia. Many cases are diagnosed annually and there is no currently available cure. Understanding the underlying disease biology of AD through the study of molecular networks, particularly by mapping clinical variants to tissue‐specific interactomes and ...

In Vitro Studies of the Prp9·Prp11·Prp21 Complex Indicate a Pathway for U2 Small Nuclear Ribonucleoprotein Activation [PDF]
Pre-mRNA splicing takes place on a large ribonucleoprotein particle, the spliceosome which contains the five small nuclear ribonucleoproteins (snRNPs), U1, U2, U4, U5, and U6. In Saccharomyces cerevisiae the mRNA splicing factors, Prp9, Prp11, and Prp21,

Different telomere functions show distinct dependence on Rap1 protein levels revealed by analyses of pre‐mRNA splicing mutants. ABSTRACT Telomeres perform multiple functions to maintain genome stability, including telomere length regulation, chromosome end protection, and meiotic chromosome dynamics.

WRAP53 Unwrapped; Roles in Nuclear Architecture and Cancer [PDF]
WRAP53 is a gene of multiple functions; it encodes for a natural antisense transcript that regulates the actions of the tumour suppressor p53, and it gives rise to a protein with oncogenic properties that is important for Cajal body formation and ...

ABSTRACT Maternally inherited transcripts are crucial for the developmental competence of salmon embryos. Due to the wide variety of broodstock conditioning approaches used in commercial aquaculture, there is a need to understand how dynamic commercial environments impact the egg transcriptome and subsequent quality.

A Role for Pre-mRNA-PROCESSING PROTEIN 40C in the Control of Growth, Development, and Stress Tolerance in Arabidopsis thaliana [PDF]
Because of their sessile nature, plants have adopted varied strategies for growing and reproducing in an ever-changing environment. Control of mRNA levels and pre-mRNA alternative splicing are key regulatory layers that contribute to adjust and ...

A Prenatal Diagnosis of Verheij Syndrome in a Fetus Harboring a de novo PUF60 Variant
Here, we report the case of a fetus affected by multiple congenital anomalies (MCA) syndrome detected during the second trimester through ultrasound scan, in which a de novo pathogenetic variant in the PUF60 gene (NM_078480.3:c.382_383del; p.Met128ValfsTer11) was identified by trio‐based exome sequencing.

Selection of alternative 5 ' splice sites: Role of U1 snRNP and models for the antagonistic effects of SF2/ASF and hnRNP A1 [PDF]
The first component known to recognize and discriminate among potential 5' splice sites (5'SSs) in pre-mRNA is the U1 snRNP. However, the relative levels of U1 snRNP binding to alternative 5'SSs do not necessarily determine the splicing outcome ...

We have purified the yeast U5 and U6 pre-mRNA splicing small nuclear ribonucleoproteins (snRNPs) by affinity chromatography and analyzed the associated polypeptides by mass spectrometry. The yeast U5 snRNP is composed of the two variants of U5 snRNA, six U5-specific proteins and the 7 proteins of the canonical Sm core.
