Short-term sustained hyperglycaemia fosters an archetypal senescence-associated secretory phenotype in endothelial cells and macrophages

open access: yesRedox Biology, 2018
Diabetic status is characterized by chronic low-grade inflammation and an increased burden of senescent cells. Recently, the senescence-associated secretory phenotype (SASP) has been suggested as a possible source of inflammatory factors in obesity ...

The multifaceted role of the SASP in atherosclerosis: from mechanisms to therapeutic opportunities

open access: yesCell & Bioscience, 2022
Background The global population of older individuals is growing, and ageing is a key risk factor for atherosclerotic cardiovascular diseases. Abnormal accumulation of senescent cells can cause potentially deleterious effects on the organism with age. As

Paracrine Senescence of Mesenchymal Stromal Cells Involves Inflammatory Cytokines and the NF-κB Pathway

open access: yesCells, 2022
It has been known that senescence-associated secretory phenotype (SASP) triggers senescence of the surrounding normal cells. However, SASP signaling regarding mesenchymal stromal cell aging remains to be fully elucidated.

Pre‐Encoded IFN‐I Sensitivity Exacerbates Memory T Cell Senescence in Solid Tumors

open access: yesAdvanced Science, EarlyView.
Type I interferon (IFN‐I) signaling promotes p21‐dependent cell cycle arrest in senescent tumor‐specific memory T cells, resulting in poor proliferative responses and solid tumor regression during cancer vaccination. Conversely, IFNα/β receptor blockade reinvigorates T cell proliferation to regress solid tumors and is more effective with increasing ...

SENOLYTICS AND SENOMORPHICS: A NEW CLASS OF DRUGS FOR ANTI-AGING THERAPY

open access: yesCentral Asian Journal of Medical Hypotheses and Ethics
Aging is a complex biological process characterized by the accumulation of senescent cells that adopt a senescence-associated secretory phenotype (SASP), releasing proinflammatory mediators that drive tissue dysfunction and the onset of age-related ...

Stochastic Nanoscale Biophysical Cues as a Basis for the Induction of Glioblastoma‐Like Transcriptional Programs in Astrocytes

open access: yesAdvanced Science, EarlyView.
Stochastic nanoscale physical cues induce glioblastoma (GBM)‐associated transcriptional traits in naïve astrocytes leading to spontaneous formation of spheroids. Cells within spheroids express activated‐MMP2 and a differential gene expression pattern involving P53 and NOTCH3, providing evidence for a role for changes in brain topography, as observed in

Extracellular vesicles, ageing, and therapeutic interventions [PDF]

open access: yes, 2018
A more comprehensive understanding of the human ageing process is required to help mitigate the increasing burden of age-related morbidities in a rapidly growing global demographic of elderly individuals.

SASP mediates chemoresistance and tumor-initiating-activity of mesothelioma cells [PDF]

open access: yesOncogene, 2011
Here we show that pemetrexed-treated mesothelioma cells undergo accelerated senescence. This is characterized by the secretion of proinflammatory and mitogenic cytokines, reminiscent of an SASP (senescence-associated secretory phenotype). Conditioned media from senescent MPM (malignant pleural mesothelioma) cells trigger the emergence of EMT ...

p16Ink4a‐Positive Hepatocytes Drive Liver Fibrosis Through Activation of LIFR Family Pathway

open access: yesAdvanced Science, EarlyView.
This study found that, following the long‐term CCl4 treatment, p16high hepatocytes appeared in zone 3, spatially co‐localizing with fibrotic areas. A specific cluster of p16high hepatocytes upregulated CTF1/LIF expression which induced HSC activation and further liver fibrosis, as revealed by single cell transcriptomic analysis.

“Comparison of in vitro models for the study of senescence of macrophages associated with a tumor

open access: yesМедицинская иммунология
Tumor-associated macrophages (TAMs) are an important and most represented population of immune cells in the tumor microenvironment. To a great extent, TAMs can determine the direction of the antitumor immune response; they can either additionally ...
