Current Management & Limitations: Traditional auditory devices lack genetic curative potential and yield variable, non‐physiological hearing outcomes. OTOF gene therapy has shown promising clinical outcomes with dual‐AAV therapy. GJB2 gene therapy remains an emerging approach with significant translational challenges. Advances in vector design and cell‐

Clock Genes Regulate the Circadian Expression of Piezo1, TRPV4, Connexin26, and VNUT in an Ex Vivo Mouse Bladder Mucosa. [PDF]
ClockΔ19/Δ19 mice is an experimental model mouse for nocturia (NOC). Using the bladder mucosa obtained from ClockΔ19/Δ19 mice, we investigated the gene expression rhythms of mechanosensory cation channels such as transient receptor potential cation ...

A Novel Striated Membrane Contact Site in Vestibular Hair Cells
The mammalian vestibular system features type I and II hair cells (HCs) with distinct neurotransmission: type I uses both ribbon synapses and rapid non‐quantal transmission (NQT). We reveal a novel striated membrane contact between type I HCs and calyceal afferents, linked to atypical mitochondria.

Extracellular Divalent Cations Regulation of Cx26 and Cx30 Hemichannels [PDF]
Due to the large size and modest selectivity of the aqueous pore, exacerbated opening of connexin hemichannels leads to loss of electrochemical gradients and of small cytoplasmic metabolites, causing cell death. Control of hemichannel opening is indispensable, and is achieved by extracellular divalent concentrations, which drastically reduces ...

ABSTRACT Hearing impairment is attributed to factors such as age, genetic predisposition, and environmental influences, among which environmental factors are considered modifiable. Among various environmental factors, the role of poor lifestyle habits is particularly critical, yet the specific mechanisms by which they contribute to hearing damage ...

Mechanisms of Cx43 and Cx26 transport to the plasma membrane and gap junction regeneration [PDF]
Previous reports have suggested that Cx26 exhibits unique intracellular transport pathways en route to the cell surface compared with other members of the connexin family. To directly examine and compare nascent and steady-state delivery of Cx43 and Cx26 to the plasma membrane and gap junction biogenesis we expressed fluorescent-protein-tagged Cx43 and

Macrophage‐Fibroblast Crosstalk Shapes Wound Repair Signaling In Vitro
Intercellular communication between macrophages and fibroblasts is critical for wound healing. Macrophages promoted fibroblast scratch closure through contact‐dependent mechanisms independent of connexins. In contrast, fibroblasts regulated macrophage cytokine secretion via connexin‐43–mediated contact signaling that increased IL10 and paracrine ...

Residues Involved in Cx26 Hemichannels Voltage Dependent Gating [PDF]
The most well-known role of Connexins (Cxs) is to communicate the cytoplasm of two adjacent cells forming a gap junction channel in the apposition zone. On the other hand, Cxs form the so called “hemichannels”, a fully functional voltage-gated non-selective ion channel, which present two gating mechanism called “slow” and “fast” gating.
