m5C methylated lncRncr3–MeCP2 interaction restricts miR124a-initiated neurogenesis

open access: yesNature Communications
AbstractCoordination of neuronal differentiation with expansion of the neuroepithelial/neural progenitor cell (NEPC/NPC) pool is essential in early brain development. Our in vitro and in vivo studies identify independent and opposing roles for two neural-specific and differentially expressed non-coding RNAs derived from the same locus: the ...

Chromatin Accessibility in Cancer: Biological Functions, Mechanisms, Therapeutic Potential, and Future Directions

open access: yesMedComm, Volume 7, Issue 3, March 2026.
Cancer remains the leading cause of mortality worldwide, and drug resistance further underscores the urgent need for innovative therapeutic strategies. Chromatin, a stable yet highly dynamic nucleoprotein complex, serves as the primary carrier of genetic material in eukaryotic cells.

RNA m5C methylation mediated by Ybx1 ensures hematopoietic stem and progenitor cell expansion

open access: yesCell Reports
Hematopoietic stem and progenitor cells (HSPCs) undergo rapid transcriptional transitions among distinct cell states and functional properties during development, but the underlying molecular mechanism is largely unknown. Here, we characterize the mRNA m5C landscape of developing HSPCs in zebrafish and found that m5C modification is essential for HSPC ...

NSUN2-mediated RNA m5C modification drives multiple myeloma progression by enhancing the stability of HIP1 mRNA

open access: yesScientific Reports
RNA 5-methylcytosine (m5C) modification is a crucial epigenetic regulation, and aberrant m5C methylation is associated with the pathogenesis of certain cancers.

The RNA m5C methylase NSUN4 protects retinal ganglion cells by regulating m5C methylation in the Hedgehog signaling pathway

open access: yes
Abstract Glaucoma, one of the leading causes of blindness worldwide, is characterized by retinal ganglion cell (RGC) damage. NOP2/Sun RNA methyltransferase 4 (NSUN4), a known 5-methylcytosine (m5C) writer, is an essential dual-function mitochondrial protein that facilitates both the process of methylation and the coordination of mitoribosome ...

NSUN2-Mediated M5c Dysregulation Promotes OSCC Ferroptosis

open access: yesInternational Dental Journal
Aim or purpose: This study aimed to investigate the role of NSUN2-mediated 5-methylcytosine (m5C) methylation in regulating ferroptosis resistance and tumor progression in oral squamous cell carcinoma (OSCC), identifying potential epitranscriptomic ...

NSUN2-mediated m5C hypermethylation of hsa_circ_0004516 promotes breast cancer brain metastasis by activating AKT signaling

open access: yesNon-coding RNA Research
Breast cancer brain metastasis (BCBM) remains fatal with elusive mechanisms. Here, we unveil the first circRNA m5C methylation landscape in BCBM through MeRIP-seq (methylated RNA immunoprecipitation next-generation sequencing) identifying 7465 BCBM ...

Elevating RNA m5C methylation provides a promising strategy for crop productivity

open access: yes
Abstract RNA 5-methylcytidine (m5C) has been identified as a key epi-transcriptomic modification of mRNAs involved in regulating multiple post-transcriptional processes. Here, we found that knockout of the RNA m5C demethylase, OsNOP2, results in elevated m5C levels and positively influences numerous agronomic traits in rice.
