Antigen processing by macroautophagy for MHC presentation [PDF]

open access: yesFrontiers in Immunology, 2011
T cells recognize antigen fragments, presented to them by MHC molecules. It lies in the interest of the immune system to display a maximal diversity of these peptides and utilize all catabolic processes to generate them.

Macroautophagy during innate immune activation [PDF]

open access: yesFrontiers in Cellular and Infection Microbiology, 2011
Innate immune activation is initiated by recognition of pathogen associated molecular patterns (PAMPs). Delivery of PAMPs to their respective receptors, regulation of receptor activity and effector functions downstream from these receptors, which ...

Macroautophagy deficiency mediates age-dependent neurodegeneration through a phospho-tau pathway [PDF]

open access: yesMolecular Neurodegeneration, 2012
Background Macroautophagy is an evolutionarily conserved mechanism for bulk intracellular degradation of proteins and organelles. Pathological studies have implicated macroautophagy defects in human neurodegenerative disorders of aging including ...

Impairment of Macroautophagy in Dopamine Neurons Has Opposing Effects on Parkinsonian Pathology and Behavior [PDF]

open access: yesCell Reports, 2019
Summary: Parkinson’s disease (PD) is characterized by the death of dopamine neurons in the substantia nigra pars compacta (SNc) and accumulation of α-synuclein.

Autophagy Flux Is Remodeled Sex- and Cell Type-Specifically During Human Aging, and Is Linked to Reduced Physical Function in Older Adults. [PDF]

open access: yesAging Cell
Autophagy‐gene transcription increases with age and is uncoupled from autophagy flux. Autophagy flux is remodeled sex‐ and cell type‐specifically and is associated with worse physical function in older adults. In a pilot intervention study, elevated PBMC autophagy flux decreased following mild exercise in older adults.

Traditional Chinese Medicine Formulas in Delaying Aging: From Theoretical Foundations to Molecular Mechanisms and Translational Perspectives. [PDF]

open access: yesJ Cell Mol Med
ABSTRACT Aging is a systemic decline in physiological integrity, driving chronic diseases such as neurodegeneration, cardiovascular disorders, and metabolic syndromes. Rapid global population aging urgently demands effective interventions. Traditional Chinese medicine (TCM) formulas, characterized by multi‐component, multi‐target, and multi‐pathway ...

Systems biology approaches for neurodegeneration and macroautophagy [PDF]

open access: yes, 2023
Neurodegenerative diseases have been identified and studied for decades but disease-modifying drugs are still unavailable for their majority. Their genetic and clinical complexity renders the identification of the precise molecular disease mechanism ...

Molecular Basis of Neuronal Autophagy in Ageing: Insights from Caenorhabditis elegans

open access: yesCells, 2021
Autophagy is an evolutionarily conserved degradation process maintaining cell homeostasis. Induction of autophagy is triggered as a response to a broad range of cellular stress conditions, such as nutrient deprivation, protein aggregation, organelle ...

Methylene Blue Induces Macroautophagy through 5' Adenosine Monophosphate-Activated Protein Kinase Pathway to Protect Neurons from Serum Deprivation

open access: yesFrontiers in Cellular Neuroscience, 2013
Methylene blue has been shown to be neuroprotective in multiple experimental neurodegenerative disease models. However, the mechanisms underlying the neuroprotective effects have not been fully elucidated.

Abberant alpha-synuclein confers toxicity to neurons in part through inhibition of chaperone-mediated autophagy. [PDF]

open access: yesPLoS ONE, 2009
BACKGROUND:The mechanisms through which aberrant alpha-synuclein (ASYN) leads to neuronal death in Parkinson's disease (PD) are uncertain. In isolated liver lysosomes, mutant ASYNs impair Chaperone Mediated Autophagy (CMA), a targeted lysosomal ...
