Mitofusin 2 Dysfunction and Disease in Mice and Men [PDF]

open access: yesFrontiers in Physiology, 2020
A causal relationship between Mitofusin (MFN) 2 gene mutations and the hereditary axonal neuropathy Charcot-Marie-Tooth disease type 2A (CMT2A) was described over 15 years ago. During the intervening period much has been learned about MFN2 functioning in

Role of mitofusin 2 in the renal stress response. [PDF]

open access: yesPLoS ONE, 2012
The role of mitofusin 2 (MFN2), a key regulator of mitochondrial morphology and function in the renal stress response is unknown. To assess its role, the MFN2 floxed gene was conditionally deleted in the kidney of mice (MFN2 cKO) by Pax2 promoter driven ...

Prognostic impact of mitofusin 2 expression in colon cancer. [PDF]

open access: yesTransl Cancer Res, 2022
Mitofusin 2 (MFN2) is involved in several biological processes, including cancer. MFN2 is downregulated in some types of cancer and inhibits cancer cell proliferation, migration, and invasion. However, the relationship between MFN2 and colon cancer remains unknown. In this study, MFN2 expression was investigated using The Cancer Genome Atlas (TCGA) and ...

Mitofusin 2 but not mitofusin 1 mediates Bcl-XL-induced mitochondrial aggregation [PDF]

open access: yesJournal of Cell Science, 2020
ABSTRACT Bcl-2 family proteins, as central players of the apoptotic program, participate in regulation of the mitochondrial network. Here, a quantitative live-cell fluorescence resonance energy transfer (FRET) two-hybrid assay was used to confirm the homo-/hetero-oligomerization of mitofusins 2 and 1 (MFN2 and MFN1), and also demonstrate

Mitofusin-2 independent juxtaposition of endoplasmic reticulum and mitochondria: an ultrastructural study. [PDF]

open access: yesPLoS ONE, 2012
Besides its role in controlling the morphology of mitochondria, mitofusin-2 has been proposed to tether mitochondria to the endoplasmic reticulum (ER), based largely on light microscopic analysis. In this study we have examined by electron microscopy the

Mitofusin 2: from functions to disease [PDF]

open access: yesCell Death & Disease, 2018
AbstractMitochondria are highly dynamic organelles whose functions are essential for cell viability. Within the cell, the mitochondrial network is continuously remodeled through the balance between fusion and fission events. Moreover, it dynamically contacts other organelles, particularly the endoplasmic reticulum, with which it enterprises an ...

Piperine Derivatives Enhance Fusion and Axonal Transport of Mitochondria by Activating Mitofusins

open access: yesChemistry, 2022
Piperine (1-piperoylpiperidine) is the major pungent component of black pepper (Piper nigrum) and exhibits a spectrum of pharmacological activities. The molecular bases for many of piperine’s biological effects are incompletely defined. We noted that the

Mitofusin-2 Regulates Platelet Mitochondria and Function. [PDF]

open access: yesCirc Res
BACKGROUND: Single-nucleotide polymorphisms linked with the rs1474868 T allele ( MFN2 [mitofusin-2] T/T) in the human mitochondrial fusion protein MFN2 gene are associated with reduced platelet MFN2 RNA expression and platelet ...

The Effect of a Novel c.820C>T (Arg274Trp) Mutation in the Mitofusin 2 Gene on Fibroblast Metabolism and Clinical Manifestation in a Patient. [PDF]

open access: yesPLoS ONE, 2017
Charcot-Marie-Tooth disease type 2A (CMT2A) is an autosomal dominant axonal peripheral neuropathy caused by mutations in the mitofusin 2 gene (MFN2).

Essential lipid autacoids rewire mitochondrial energy efficiency in metabolic dysfunction‐associated fatty liver disease

open access: yesHepatology, EarlyView., 2022
Increased liver content of DHA‐derived small lipid autacoids (i.e resolvin D1 and maresin 1) associates with enhanced mitochondrial oxidative phosphorylation, fatty acid β‐oxidation and bioenergetic metabolic flux. These features provide hepatic protection from steatotic, pro‐inflammatory and fibrogenic insults.
