Protoplast Isolation and Transfection in Wheat

2022
Wheat is one of the major staple crops around the world. A transient expression system is crucial for gene functional studies in wheat as stable transfection is still difficult in most cultivars. Protoplasts could serve as a versatile transient expression tool in wheat research.

Fusion of Isolated Plant Protoplasts

Nature, 1970
A first step towards somatic hybridization in plants has been achieved. Protoplasts have been isolated and seen to fuse.

The isolation of barley-aleurone protoplasts

Planta, 1971
A procedure is described for the isolation of large numbers of viable aleurone protoplasts. After treatment with "Onozuka" cellulase protoplasts were obtained which were surrounded by a thin, "Onozuka"-resistant wall. These cells were termed "spheroplasts".

Reuse of enzymes for isolation of protoplasts

Plant Cell Reports, 1985
A method has been developed for the reuse of cell wall digesting enzymes to isolate protoplasts from actively-growing suspension cultures of plant cells. Protoplasts could be satisfactorily prepared as many as three times using the same enzyme mixture without any loss in yield or viability of the isolated protoplasts. The yields of nuclei isolated from

Protoplast Isolation and Staining

2015
The successful isolation of mesophyll protoplasts from plant species has become a versatile tool for in vivo imaging of subcellular structures. Taking advantage of the various cytochemical probes available, the subcellular localization of specific organelles can be visualized in live protoplasts.

Isolation of Protoplasts

1992
Although protoplasts have been isolated from a range of somatic tissues and successfully cultured, the success in isolating the protoplasts from reproductive tissues has been rather limited. This is basically due to technical difficulties. In the pollen, for example, the exine, which is made up of sporopollenin, is resistant to enzymatic degradation ...

Isolation and regeneration of protoplasts from Ectocarpus.

Cold Spring Harbor protocols, 2012
This article describes how to obtain isolated cells with no surrounding cell wall by enzymatic digestion of Ectocarpus filaments. The resultant protoplasts are totipotent and regenerate to produce individual algae under appropriate culture conditions.

ISOLATION OF SUNFLOWER PROTOPLASTS

The isolation, regeneration and fusion of Phycomyces protoplasts

Molecular and General Genetics MGG, 1974
Germlings of Phycomyces blakesleeanus were converted to protoplasts using an enzyme solution of chitinase and chitosanase. The protoplasts were regenerated to normal growing mycelia. Protoplasts of different auxotrophic strains fused in diluted natural seawater and in Ca(NO3)2 solutions. 36 prototrophic strains were isolated (Table 1).

Isolation and Transfection of Maize Endosperm Protoplasts

2022
Endosperm of cereal crops is the main component of its grain. Improvement of endosperm traits will bolster grain yield and quality. Functional analysis of endosperm trait-related genes often requires the use of an endosperm cell system. Here, we describe a protocol for the isolation and transfection of maize endosperm cell protoplast.
