Senotherapeutics to Counteract Senescent Cells Are Prominent Topics in the Context of Anti-Ageing Strategies [PDF]
Cellular senescence is implicated in ageing and associated with a broad spectrum of agerelated diseases. Importantly, a cell can initiate the senescence program irrespective of the organism’s age.

Senolytics and exercise: Dual modalities for rejuvenating muscle
Abstract figure legend The role of senolytics on the heart and skeletal muscle. Senescent cell burden increases with ageing, disuse and disease. The senolytics dasatinib+quercetin (D+Q), navitoclax and fisetin, as well as exercise, eliminate senescent cells, reducing senescent cell burden and their senescence‐associated secretory phenotype (SASP ...

Repurposing previously approved drugs may fast track the route to the clinic for potential senotherapeutics and improves the inefficiency of the clinical drug development pipeline. We performed a repurposing screen of 240 clinically approved molecules in human primary dermal fibroblasts for their effects on CDKN2A expression.

Background: Senomorphics have been considered as an effective alternative paradigm of senotherapeutics to regulate cellular senescence and related disorders by inhibiting the deleterious effects of senescence-associated secretory phenotype (SASP) components secreted by senescent cells without inducing cell death.

The Quest for Eternal Youth: Hallmarks of Aging and Rejuvenating Therapeutic Strategies
The impressive achievements made in the last century in extending the lifespan have led to a significant growth rate of elderly individuals in populations across the world and an exponential increase in the incidence of age-related conditions such as ...

Uncovering a new layer of translational control, this study reveals how TRMT6/TRMT61A‐mediated tRNA‐m1A methylation drives pro‐tumorigenic senescence in colorectal cancer. By selectively enhancing ARG2 translation, this epitranscriptomic axis triggers an NF‐κB‐dependent SASP.

Senescence of skeletal stem cells and their contribution to age-related bone loss [PDF]
Human aging is linked to bone loss, resulting in bone fragility and an increased risk of fractures. This is primarily due to an age-related decline in the function of bone-forming osteoblastic cells and accelerated cellular senescence within the bone ...

A mechanically robust, ROS‐responsive hydrogel is engineered to treat intervertebral disc degeneration by targeting the NF‐κB–LCN2 inflammatory feedback loop. By delivering wedelolactone specifically within oxidative niches, this system disrupts macrophage–disc cell crosstalk.

Senescent AECⅡ and the implication for idiopathic pulmonary fibrosis treatment
Idiopathic pulmonary fibrosis (IPF) is a chronic and lethal lung disease with limited treatment options. The onset of IPF increases with age, indicating that aging is a major risk factor for IPF.

Engineering Immune Cell to Counteract Aging and Aging‐Associated Diseases
This review highlights a paradigm shift in which advanced immune cell therapies, initially developed for cancer, are now being harnessed to combat aging. By engineering immune cells to selectively clear senescent cells and remodel pro‐inflammatory tissue microenvironments, these strategies offer a novel and powerful approach to delay age‐related ...
